You can think value and say Action Comics #1 but really think about a comic that would mean a lot to you (and I bet Action #1 would mean a lot to a lot of comic fans) - but what's the one comic, if your Fairy Godparent showed up and offered it to you, that you'd want?
Me? Teenage Mutant Ninja Turtles #1 (1984), first printing of course. Ninja Turtles changed my life - and I mean this "pre cartoon" version. It taught me that I can make comics myself, and I can make up the characters no matter how silly and pull it off if I do it right. I never viewed the art of comic making the same again (or really anything else - my entire DIY personality goes back to TMNT).

Easy - Fantastic Four #1.
I've been an FF fan since I was 10. It wouldn't even have to be anywhere near perfect. Something with an attached cover, no major folds or tears, and in generally clean condition and I'd be thrilled beyond belief. Aw who am I kidding... I'd take just about any condition of FF #1.

Detective Comics #27 for me. First appearance of the Bat. This is what I was going to say. First printing, and signed. I would have it framed above my bed forever and always. Also: Saga #1 first printing. I didn't jump on until the second trade was out, so it would be AWESOME to have the first issue.
